Amyloplasts from Solanum tuberosum (potato tuber)

The SEM image shows amyloplasts from a high moisture content (72 – 82%) potato tuber (Solanum tuberosum). Prepared using the Quorum PP3010 cryo-preparation chamber. Preparation involved cryo-immobilization at -207 °C in slush nitrogen, fracturing at -140 °C, and sputtering with 4 – 6 nm of Iridium. A sublimation period of 6 min was used for the tuber specimen to reveal both the amyloplasts and cellulose cell wall compartments. Imaged on Tescan Amber.
