Pollen from pumpkin stamen

The SEM image shows pollen from a pumpkin stamen prepared using the Quorum PP3010 preparation chamber. A section of stamen was taken and mounted using mounting media (50:50 mixture of Tissue-Tek OCT (Optimal Cutting Temperature) compound and colloidal graphite), vitrified in slushed nitrogen (LN2) at 207 °C, fracturing at -140 °C, sublimed to -90°C for 2 min and sputter coated with 4– 7 nm of Iridium. Imaged on Tescan Amber.
